a) Order your alkyne-modified oligo by sending an email to oligo@mymetabion.com.
Make sure to include all relevant information like
- shipping address & contact information (person, phone, fax, email)
- billing address & contact information (person/customer ID, phone, fax, email, VAT #)
- quotation and/or PO # if any
- product specification including
- oligo name
- purification option - standard or HPLC
- synthesis scale/desired minimum final quantity delivered
- delivery form (dry or wet)
- sequence 5´-3´ and position of the Alkyne-modification according to the following instructions:
Use respective Modification shortcuts listed in the table below.
There are 2 ways to introduce Alkyne groups to the 5´end of the oligo:
1) Alxyl is sequence independent, adding an Alkyne group via a C6 (hexynyl)- linker to the phosphate of the 5´ nucleoside.
2) AldU and AldC carry a C8-Alkyne motif at the C5 position of the respective Pyrimidine structure of either dU (replacing dT) or dC. Hence, if there is a C or T at the 5´ end of the oligo sequence, Alkyne-activation can also be introduced through AldC or AldU (sequence dependent).
For internal Alkynylation either choose AldU or AldC to replace dT and/or dC within the oligo sequence.
For Alkyne-activation of the 3´ end again, there are two options:
1) Alrol is sequence independent, adding an Alkyne group via a serinol-linker to the 3´ OH of the 3´ nucleoside (3´OH-blocking).
2) AldU and AldC carry a C8-Alkyne motif at the C5 position of the respective Pyrimidine structure of either dU (replacing dT) or dC. Hence, if there is a C or T at the 3´ end of the oligo sequence, Alkyne-activation can also be introduced through AldC or AldU (sequence dependent) without blocking the 3´OH end.

“Click”-chemistry allows for simple, quick, and efficient post-synthetic modification at multiple positions of alkynylated DNA (e.g. for FISH, EMSA etc.). Moreover, alkynylated DNA itself can be used directly in various applications including nanotechnology.
