Description
mi-Blood Genomic DNA Kit is designed for the rapid, simultaneous preparation of highly pure genomic DNA from whole blood, serum, plasma, or other body fluids. The obtained DNA can be used directly for PCR, Southern Blotting, or any kind of enzymatic reaction.
The kit allows purification of up to 10 µg of pure genomic DNA from 200 µl whole blood with an A260/280 ratio between 1.6 and 1.9 and a typical concentration of 20-100 ng per µl.
Blood treated either with EDTA, citrate, or heparin can be used. The procedure is optimized for a sample volume of 200ul and samples can be diluted with sterile PBS buffer.
Mammalian whole blood genomic DNA prepared with this kit is suitable for use as a template in PCR and Southern Hybridization.
Principle
The overall principle of DNA purification by silica-membrane technology is the specific and selective adsorption of DNA molecules to the silica-gel–membrane in the spin column. The cells are lysed during a short incubation with Proteinase K in the presence of chaotropic salt which immediately inactives all nucleases and enhances the binding of DNA. After the binding of the DNA in optimized high-salt buffers to the surface, all contaminants like primers, nucleotides, enzymes, mineral oil, salts, agarose, ethidium bromide, and other impurities from DNA samples, can be removed through specialized wash buffers. The purified DNA can be eluted by low-salt buffer or distilled water.
