metabion, the expert in custom synthesis of primers and probes, now offers 2x ready-to-use real-time PCR hot start mixes using either green-fluorescent DNA stain or labeled DNA probes. The reaction chemistry of the mixes is optimized for block-based PCR instruments (no glass capillaries) and suitable for most qPCR machines and platforms (+/- ROX reference).
Master mixes contain all reagents (except template and oligos) required for qPCR in a premixed 2x concentrated ready-to-use solution. High specificity and sensitivity of the mixes are achieved by an optimized hot-start polymerase. Its activity is blocked at ambient temperature and switched on automatically at the onset of the initial denaturation. The thermal activation prevents the extension of nonspecifically annealed primers and primer-dimer formations at low temperatures during PCR setup.
The mixes contain dUTP instead of dTTP in order to eliminate carry-over contamination of DNA from previous PCR reactions by the use of UNG (Uracil-N-Glycosylase).
UNG (Uracil-N-Glycosylase)
The UNG treatment at the onset of thermal cycling removes Uracil residues from dU-containing DNA of previous PCR amplifications preventing it from serving as template (carry-over contamination control).
ROX passive reference dye
ROX is a passive reference dye and does not take part in the PCR reaction. However, it allows to normalize for non-PCR related signal variation and provides a baseline in multiplex reactions.

EvaGreen® Master Mixes
mi-real-time EvaGreen® Master mixes (2x conc.) are designed for the quantitative real-time analysis of DNA samples using the fluorescent DNA stain EvaGreen®. The fluorescent dye in the master mix intercalates into the amplification product during the PCR process enabling for rapid analysis of target DNA. No need for sequence-specific labeled probes. It provides an easy-to-handle and powerful tool for quantification of sample DNA in a broad dynamic range of up to 6 orders of magnitude with exceptional sensitivity and precision.
EvaGreen® is superior to the widely used SYBR®Green and is suitable for precise HRM analysis (High resolution melting).

Probe Master Mixes
mi-Real-Time Probe Master Mixes (2x conc.) are designed for quantitative real-time analysis of DNA samples using DNA probe based detection. Master Mixes are recommended for 5´Nuclease- or FRET Hybridization Probe-assays using fluorescent probes. Being the expert in custom synthesis of primers and probes for real-time PCR, we now also offer Probe Master Mixes as valuable high quality “accessories” for the successful performance of your assays.
